Esketamine, the main compound in Spravato, works differently than traditional SSRI or SNRI antidepressants, making it a worthwhile treatment option for those with TRD. Spravato targets the brain’s glutamate receptors, which regulate mood and cognitive function. It also boosts neuroplasticity—the brain’s ability to adapt and form new connections—potentially healing damage caused by chronic depression.

While oral medications are effective for some, one in three people experience little to no improvement. Often, people must navigate unpleasant side effects that make antidepressants intolerable. Current research shows that 88 percent of patients experienced a significant reduction in symptoms after one Spravato treatment.

What To Expect During Treatments

Spravato is delivered in phases. During the first four weeks (induction), you will come to our office twice per week. In weeks five through eight, dosing typically occurs once per week to stabilize your response. After week eight, we transition to the maintenance phase—generally every other week and adjusted as needed based on your progress.

Studies show that patients who continued to use Spravato at least every other week after completing their induction and maintenance phases were less likely to experience a relapse in depression symptoms compared to those who stopped treatments.

While many patients experience an improved mood after the first 24 hours of Spravato use, you should feel significant symptom reduction after the first four weeks of treatment.
